Mass Spectrometry for Proteins

Mass Spectrometry Section of the Collaborative Protein Technology Resource (Bldg. 37)

Core Capabilities:

  • Identification of proteins in complexes, organelles, subcellular fractions, or fluids.
  • Global relative protein quantitation.
  • Quantitation by isotopic labeling of cells in culture (SILAC) or by reaction with isobaric tags (iTRAQ/TMT) for analysis of proteins from mouse or clinical samples.
  • Quantitation can be multiplexed, allowing comparison of multiple conditions/states/treatments at once.
  • Label-free relative quantitation.
  • Absolute quantitation of specific proteins in a complex mixture.
  • In a targeted approach, absolute quantitation of the level of a particular protein in a complex mixture (e.g., serum, conditioned media, lysate).
  • Analysis of post-translational modifications.
  • Identification of the site of modification for phosphorylation, acetylation, ubiquitylation, methylation, etc.
  • Relative quantitation of changes in global modification patterns.

Protein Characterization Laboratory of the Cancer Research Technology Program (CRTP): Mass Spectrometry Center

The Protein Characterization Lab (PCL) is equipped with the latest in mass spectrometry (MS) instrumentation for proteomic and metabolomic analysis. Areas of expertise include protein complex characterization, global and target protein quantitation, and identification of post-translational modifications. The Center also provides educational and training opportunities for scientists on the use, interpretation, and applications of MS technologies.

Consultation and collaboration for Protein-Protein Interactions (PPI) studies:

The PCL provides a complete service protein complex characterization pipeline.  This service works in conjunction with the Protein Expression Laboratory (PEL), who clone the protein of interest into an expression vector.  The vector is transfected and expressed in an appropriate cell line followed by immunoprecipitation of the protein complex and analysis using mass spectrometry.
